Are there any bots for single player LOD 1.10 that will run Countess? I'm new to the whole bot concept, so any help/advice here would be appreciated. Thanks!

D2JSP will run on other multiplayer which is single player but on the internet.

Most bots worth by faking packets to the server. The only suggestion I have for you are autoit bots... which will be unable to run countess. But since you are using single player why don't you just use a char editor to get the items you want? Surely this will be a far less time consuming process to get the items/runes you want.
